Ii. Bioavailability and in Vivo Formation of Aa-2g

چکیده
I . INTRODUCTION It is well known that L-ascorbic acid (AsA) has multifarious functions in our body. Humans who are unable to synthesize AsA in vivo have to absorb it as an essential nutrient from foods and some supplements. However, AsA is easily oxidized by oxygen in aqueous solutions, followed by oxidative degradation. To improve such instability, some derivatives including monoor di-substituted AsA have been synthesized chemically. Recently, we have found a new AsA conjugate which is effectively synthesized by enzymatic transglucosylation. This compound is characterized by its high stability in aqueous solutions and its non-reducibility. Since enzymatic process surpasses chemical process with regard to its selectivity, efficiency and cost, the stable AsA glucoside has a bright prospect of developing the new field of applications as AsA supplements. In this paper we describe its enzymatic formation, property, bioavailability and biological activity.

2-O-α-d-Glucopyranosyl-l-ascorbic acid (AA-2G) is one of the stable ascorbic acid (AA) derivatives known as provitamin C agents. We have previously synthesized two types of monoacylated derivatives of AA-2G, 6-O-acyl-2-O-α-d-glucopyranosyl-l-ascorbic acids having a straight-acyl chain of varying length from C₄ to C18 (6-sAcyl-AA-2G) and a branched-acyl chain of varying length from C₆ to C16 (6-...
متن کاملTransdermal Delivery of Insulin by Biodegradable Chitosan Nanoparticles: Exvivo and In vivo Studies
Insulin-loaded biodegradable chitosan nanoparticle was prepared by the polyelec-trolyte complex formation method. The prepared nanoparticles were in the size of 110 nm and had high entrapment (91.0%) capacity. The transdermal nanoinsulin was characterized by in vivo hypoglycemic effects. Plasma glucose was decreased to the range of 80.34 to 96.74 mg/dl, and insulin levels were in...
متن کامل2-O-α-D-Glucopyranosyl-l-ascorbic acid as an antitumor agent for infusion therapy
Ascorbic acid (AA) has been reported as a treatment for cancer patients. Intravenous (iv) administration of high-dose AA increases plasma AA levels to pharmacologic concentrations and generates reactive oxygen species (ROS) to exert anti-tumor activity via enhancement of oxidative stress. However, AA is very unstable in aqueous solutions and it is impossible to preserve AA for a long period in ...
متن کاملIn vitro assessment of iron availability from commercial Young Child Formulae supplemented with prebiotics
PURPOSE Iron is essential for development and growth in young children; unfortunately, iron deficiency (ID) is a significant public health problem in this population. Young Child Formulae (YCF), milk-derived products fortified with iron and ascorbic acid (AA, an enhancer of iron absorption) may be good sources of iron to help prevent ID. Furthermore, some YCF are supplemented with prebiotics, n...
متن کاملPotential Antitumor Activity of 2-O-α-d-Glucopyranosyl-6-O-(2-Pentylheptanoyl)-l-Ascorbic Acid
Intravenous administration of high-dose ascorbic acid (AA) has been reported as a treatment for cancer patients. However, cancer patients with renal failure cannot receive this therapy because high-dose AA infusion can have side effects. To solve this problem, we evaluated the antitumor activity of a lipophilic stable AA derivative, 2-O-α-d-glucopyranosyl-6-O-(2-pentylheptanoyl)-l-ascorbic acid...
